目的:筛选出产高活力糖化酶的优良菌株;方法:经过紫外线与光复活交替处理单孢子悬液,利用透明圈法和摇瓶发酵进行初筛和复筛;结论:筛选出一株菌株12,通过摇瓶对照实验发现,其酶活达到了5544.3U/mL,比出发菌株提高了25.41%,效果较显著,选育得到的菌株经遗传稳定性试验表明有较好的遗传稳定性。

Aim: To isolate high activity saccharifying enzyme.
Methods: The spore suspension were alternately performed by UV and photoreactivation, and the high-producing strain was primary screened by the mothod of transparent circle on plate. The secondary screening was carried out by the flask culturation.
Conclusion: A strain 12 was obtained by UV mutagenesis. Using flask culturation, the enzyme activity was 5544.3 U/mL, which was 25.41% higher than of control strain, and the effect was significant. The mutanted strain identified by the test of genetic stability showed favorable stability.
