Chromatographic analysis were carried out on a high-performance liquid chromatograph (Dionex Corporation, Sunnyvale, CA,USA) equipped with a P680 HPLC Pump, Thermostatted Column Compartment TCC-100 and a Dionex Photodiode Array DAD-100 Detector (DAD). The HPLC column was TSK gel ODS-80Ts QA(150 mm 4.6 mm i.d.) (Tosoh, Tokyo, Japan) and was protected with a Transgenomic CARB Sep Coregel 87C Guard Cartridge(Transgenomic Incorporation, Omaha, NE, USA). The temperature was set at 35 8C. Extracts were passed through a 0.45 mm pore syringe filter from Anpel Corporation (Shanghai, China) and a volume of 10 mL solution was injected for HPLC.Flow rate was 0.8 mL min1 and the mobile phase consisted of 1.5% phosphoric acid water solution as solvent A (1.5:98.5; v/v, H3PO4:H2O) and phosphoric acid/formic acid/acetonitrile/water (1.5:20:25:53.5; v/v, H3PO4:HCOOH:CH3CN:H2O) as solvent B (Sakata et al., 1995). The linear gradient profile was 20% B at 0 min, 70% B at 80 min, and then returned to 20% B in 10 min. Data were recorded on a computer with the Chromeleon software (Dionex Corporation,Sunnyvale, CA, USA). Chromatograms were acquired at 515 and 340 nm and photodiode array spectra were recorded between 200 and 800 nm. Anthocyanins were primarily identified according to the retention time and UV–vis spectral chromatography by comparison with authentic samples and published data (Wang et al., 2001b; Zhang et al., 2007).
Each peak was identified by HPLC–ESI-MSn (Agilent Technologies,Palo Alto, CA, USA). The interface was an API-electrospray(Agilent 1100 LC/MSD Trap VL) operating in positive ionization mode monitoring of the protonated molecular ions at the following operating conditions: gas (N2) temperature, 350 8C; flow rate,6.0 L min1; nebulizer pressure, 241.3 kPa; octopole RF amplitude,150 Vpp; skim 1 voltage, 45.5 V; skim 2 voltage, 6.0 V; capillary exit, 123.9 V and cap exit offset, 78.4 V. The temperature was set at 35 8C. The flow rate was 0.8 mL min1 and the injection volume was 5 mL. The mobile phase was made up of solvent A (0.1%trifluoroacetic acid in water) and solvent B (acetonitrile). The gradient was 5% B at 0 min, 30% B at 40 min, and then returned to 5% B in 10 min. The ion trap mass spectrometer scanned from m/z 100 to 800. The LC/MSD Trap software version 5.2 provided complete control over all instrumentation (Fu et al., 2006).Anthocyanins were accurately identified based on comparison of the mass spectrometric data with those of authentic standards,references, and some guidelines (Wu and Prior, 2005a,b).
